Abstract
[Objective] To investigate the effects of Huangyuan moxibustion on mitochondrial energy metabolism in rats with spleen deficiency-type chronic fatigue syndrome(CFS)and to clarify its mechanism of action. [Methods] Forty SPFgrade male SD rats were randomly divided into five groups after one week of adaptive feeding:normal group, model group, herbal cake group(Huangyuan moxibustion group),suspended moxibustion group, and starch cake group, with 8 rats in each group. A spleen deficiency-type CFS model was established by chronic restraint, exhaustive swimming, and high-lactose diet feeding. Moxibustion was applied at Guanyuan(CV4)acupoint. The herbal cake group received moxibustion with a herbal cake composed of Codonopsis Radix,Poria,Zingiberis Rhizoma, and Glycyrrhizae Radix et Rhizoma Praeparata(3∶2∶2∶2). The suspended moxibustion group received mild suspended moxibustion. The starch cake group received moxibustion with a starch cake. After 5 days of intervention, the following indicators were measured:general condition score, body weight change, exhaustive swimming time, tail suspension test data,Morris water maze data,ATP content and Na+/K+-ATPase and Ca2+/Mg2+-ATPase activities in liver, small intestine, skeletal muscle, and hippocampus tissues, and mitochondrial morphology observed by transmission electron microscopy. [Results] The herbal cake group showed significantly better general condition scores than the model group and suspended moxibustion group(P<0.01),with more pronounced weight gain(P<0.01). The average exhaustive swimming time was longest(5.45 min)and the average immobility time in the tail suspension test was shortest(25.62 s)in the herbal cake group, with exercise endurance approaching that of the normal group. In the Morris water maze, the herbal cake group showed significantly faster swimming speed than the starch cake group and suspended moxibustion group(P<0.05). The Ca2+/Mg2+-ATPase activity in liver tissue of the herbal cake group was significantly higher than that of the model group(P=0.049),while it was lower than the model group in small intestine, skeletal muscle, and hippocampus tissues. Under electron microscopy, mitochondrial structural damage in the herbal cake group was significantly improved, and autophagosomes(double membrane structures containing cristae fragments)were observed. [Conclusion] Huangyuan moxibustion showed a tendency to be superior to suspended moxibustion and starch cake moxibustion in improving the overall status and some energy metabolism indicators in rats with spleen deficiencytype CFS. Huangyuan moxibustion may treat spleen deficiency type CFS by bidirectionally regulating Ca2+/Mg2+ ATPase activity and activating mitophagy to repair mitochondrial structural damage and improve energy metabolism disorders.
