疏风通络方对哮喘大鼠嗜酸细胞跨膜迁移 相关因子VCAM-1/PI3K/Rac-1/NOX2/ NOX4/SHP-2表达影响的研究

摘要:
[目的] 探讨疏风通络方对哮喘大鼠嗜酸细胞跨膜迁移的影响。[方法] 选择SPF级雄性Wistar大鼠70只为研究对象, 随机分为正常组、模型组、地塞米松组、DPI组、中药低、中、高剂量组, 共7组, 每组大鼠10只。第1天致敏:正常组给予生理盐水1 mL腹腔注射致敏, 而其他6组给予1 mL造模液(含V级卵清蛋白100 mg,氢氧化铝100 mg和灭活百日咳杆菌6×109个)腹腔注射致敏;第15天开始激发:将各组大鼠分别置于相同大小的雾化箱内, 正常组给予生理盐水6 mL雾化激发, 其他6组给予5%的 V级卵清蛋白溶液6 mL雾化激发, 每次激发前2 h正常组给予生理盐水1 mL灌胃, 而其他6组给予相应的药物灌胃。各组每天激发1次, 每次激发30 min,连续激发10 d后处死大鼠, 并采集相应标本, 并进一步检测影响嗜酸细胞跨膜迁移的相关指标。[结果] 模型组肺组织嗜酸细胞跨膜迁移相关调节因子血管内皮黏附分子-1(VCAM-1)、磷脂酰肌醇3激酶(PI3K)、Rac-1、NADPH氧化酶2(NOX2)、NADPH氧化酶4(NOX4)、酪氨酸磷酸酶(SHP-2)出现高表达, 高于正常组(P<0.001);各治疗组与模型组相比, 地塞米松组抑制上述因子表达的效果最好(P<0.001),中药高剂量组的抑制效果略差于地塞米松组, 但两者差异无统计学意义(P>0.05),氯化二联苯碘(DPI)组、中药低、中剂量组抑制效果一般。[结论] 疏风通络方对哮喘大鼠嗜酸细胞跨膜迁移有一定的阻抑作用, 这可能是其平喘作用机制之一。

Influence of Shufeng Tongluo prescription on expression of VCAM-1/PI3K/Rac-1/NOX2/NOX4/SHP-2 related factors in eosinophils transendothelia migration in lung of rats with asthma

Abstract:
[Objective] To explore the effect of Shufeng Tongluo decoction on eosinophils transendothelia migration in lung of rats with asthma. [Methods] Seventy Wistar rats (SPF) were randomly divided into seven groups: normal group, model group, dexamethasone group, DPI group, low-dose group of Chinese medicine, medium-dose group of Chinese medicine, and high-dose group of Chinese medicine with 10 rats in each group. The rats in normal group were sensitized by 1 mL of intra-peritoneal injection of normal saline. The other 6 groups were given with 1 mL sensitization liquid, contained 100 mg OVA (Grade V), 100 mg aluminum hydroxide and 6×109 inactivated whooping cough bacili on the first day of experimental study. From 15th day, the rats in normal group were administrated with 1 mL of normal saline orally, 2 hours later the rats were challenged by 6 mL normal saline in a box of aerosol inhalation once a day for 30 min a time and keeping for 10 days; the rats in other 6 group were respectively administrated with corresponding drugs orally and 2 hours later were challenged by 6 mL OVA (5%, V Grade) of aerosol inhalation once a day, for 30 min a time and keeping for 10 days, And 10 days later all rats were killed. The samples from rats were collected for examination. [Results] The expression of VCAM-1/PI3K/Rac-1/NOX2/NOX4/SHP-2 related factors to eosinophils transendothelia migration in lung of rat with asthma were significantly higher than that of normal group rats(P<0.001). Compared with the model group, dexamethasone group could significantly inhibit the expression of above factors(P<0.001). The effect of high-dose group of Chinese medicine was similar to dexamethasone group(P>0.05), the other treatment groups (DPI group, low-dose group of Chinese medicine and medium-dose group of Chinese medicine ) only had general inhibition effect. [Conclusion] Shufeng Tongluo decoction has some certain suppression effect on eosinophils transendothelia migration in lung of rats with asthma to a certain extent. It may be one of its action mechanisms in controlling asthma.
