| Abstract

| [Objective] To observe the effects of Tongxie Yaofang and Sini Powder on PI3K/AKT/GSK-3β signal pathway and insulin resistance in KK-Ay mice.[Methods] Male 6 to 8 week old KK-Ay mice were used to establish a diabetes model. They were randomly divided into model group,and low,medium,high dosage groups of Chinese medicine. C57BL/6J mice of the same week were taken as the normal group. After 4 weeks of intervention,fasting insulin levels,insulin and islet function of mice were detected,and the protein phosphorylation and mRNA expression of PI3K,AKT and GSK-3β in mice liver were detected by real time PCR and Western blot.[Results] Compared with the normal group,the fasting blood glucose and insulin resistance index of the model group mice were significantly increased (P<0.05),while the phosphorylated protein levels and gene expression levels of PI3K,AKT,GSK-3β were significantly reduced (P<0.05). Compared with the model group,the fasting blood glucose and insulin resistance index in the high dose group were significantly reduced (P<0.05),while the phosphorylated protein levels and gene expression levels of PI3K,AKT,GSK-3β were significantly increased (P<0.05).[Conclusion] Tongxie Yaofang and Sini Powder may improve insulin resistance in KK-Ay mice by affecting PI3K/AKT/GSK-3β signaling pathway. |
