| Objective To explore whether acupuncture treatment can regulate the SIRT1/PGC-1α pathway and affect the pain threshold in rats with myofascial pain syndrome (MPS). Methods SD rats were separated into the Con group, the MPS group, the non-invasive and non-acupoint control group, the acupuncture group, the acupuncture +EX527 (Sirt1 inhibitor) group and the celecoxib group. The MPS model was established by combining striking with centrifugal motion. The thermal withdrawal latency and the spontaneous electricity activity of the left lower limb of rats were detected. ELISA was used to measure the levels of TNF-α, IL-6 and IL -1β in serum and PGE2, SP and 5-HT in spinal cord tissue. HE staining was implemented to measure the pathological morphological changes of muscle tissue. The positive expression of GFAP and OX-42 in spinal cord tissue was observed by immunofluorescence. The expression levels of SIRT1 and PGC-1α proteins in spinal cord tissue were detected by Western blot. Results Compared with the Con group and the non-invasive and non-acupoint control group, the muscle tissue of rats in the MPS group was clearly damaged, the TWL value, and the protein expression levels of SIRT1 and PGC-1α in the spinal cord tissue were lower, while the frequency of spontaneous potential activity, the levels of serum TNF-α, IL-6 and IL-1β, the levels of PGE2, SP and 5-HT in spinal cord tissue, and the numbers of GFAP and OX-42 positive cells were higher (P < 0.05). Compared with the MPS group and the non-invasive and non-acupoint control group, the muscle tissue morphology of rats in the acupuncture group and the celecoxib group was clearly improved, the TWL value, and the protein expression levels of SIRT1 and PGC-1α in the spinal cord tissue were higher, while the frequency of spontaneous potential activity, the levels of serum TNF-α, IL-6 and IL-1β, the levels of PGE2, SP and 5-HT in spinal cord tissue, and the numbers of GFAP and OX-42 positive cells were lower (P < 0.05). EX527 could weaken the improvement effect of acupuncture treatment on the pain threshold of MPS rats (P < 0.05). The various detection indicators of rats in the acupuncture group and the celecoxib group were at the same level (P > 0.05). Conclusion Acupuncture treatment may reduce inflammation in MPS rats, increase the pain threshold and improve MPS by activating the SIRT1/PGC-1α pathway. |
